GHRP-2 vs MK-677 (Ibutamoren)

An educational, source-based comparison of GHRP-2 and MK-677 (Ibutamoren) — how each peptide works, what it's researched for, and what to know before going deeper.

Growth Hormone Axis
GHRP-2

Ghrelin-mimetic GH secretagogue with appetite effects.

Mechanism

A synthetic hexapeptide that activates the ghrelin/GHS-R1a receptor, stimulating endogenous GH release. Compared to Ipamorelin, GHRP-2 produces a stronger GH pulse but also modestly elevates cortisol, prolactin, and appetite.

Research areas
  • GH release in adult GH deficiency
  • Appetite stimulation in cachexia models
  • Diagnostic GH stimulation testing
Considerations
  • Not FDA-approved.
  • Less receptor-selective than Ipamorelin; expect appetite and cortisol effects.
  • Prohibited by WADA.

Growth Hormone Axis · Small Molecule
MK-677 (Ibutamoren)

Orally bioavailable non-peptide ghrelin mimetic.

Mechanism

Technically a small-molecule ghrelin receptor agonist (not a peptide), often grouped with peptide GH research. Orally bioavailable, it elevates GH and IGF-1 over 24 hours and increases appetite.

Research areas
  • GH/IGF-1 elevation in adults
  • Lean mass and bone density in elderly
  • Sleep quality (slow-wave sleep)
Considerations
  • Not FDA-approved.
  • Insulin resistance and fluid retention reported.
  • Banned by WADA.
